ABSTRACT

Organo-mineral fertilizers supplemented with biological additives are an alternative to chemical fertilizers. In this study, thermoresistant microorganisms from composting mass were isolated by two-step procedures. First, samples taken at different time points and temperatures (33 days at 52 ºC, 60 days at 63 ºC, and over 365 days at 26 ºC) were pre-incubated at 80 oC for 30 minutes. Second, the microbial selection by in vitro culture-based methods and heat shock at 60 oC and 100 oC for 2h and 4h. Forty-one isolates were able to grow at 60 °C for 4h; twenty-seven at 100 °C for 2h, and two at 100 °C for 4h. The molecular identification by partial sequencing of the 16S ribosomal gene using universal primers revealed that thirty-five isolates were from eight Bacillus species, one Brevibacillus borstelensis, three Streptomyces thermogriseus, and two fungi (Thermomyces lanuginosus and T. dupontii). Data from amylase, phytase, and cellulase activity assays and the enzymatic index (EI) showed that 38 of 41 thermo-resistant isolates produce at least one enzyme. For amylase activity, the highest EI value was observed in Bacillus licheniformis (isolate 21C2, EI= 4.11), followed by Brevibacillus borstelensis (isolate 6C2, EI= 3.66), Bacillus cereus (isolate 18C2, EI= 3.52), and Bacillus paralicheniformis (isolate 20C2, EI= 3.34). For phytase, the highest EI values were observed for Bacillus cereus (isolate 18C2, EI= 2.30) and Bacillus licheniformis (isolate 3C1, EI= 2.15). Concerning cellulose production, B. altitudinis (isolate 6C1) was the most efficient (EI= 6.40), followed by three Bacillus subtilis (isolates 9C1, 16C2, and 19C2) with EI values of 5.66, 5.84, and 5.88, respectively, and one B. pumilus (isolate 27C2, EI= 5.78). The selected microorganisms are potentially useful as a biological additive in organo-mineral fertilizers and other biotechnological processes.


Os fertilizantes organo-minerais suplementados com aditivos biológicos são uma alternativa aos adubos químicos. Neste estudo, microrganismos termoresistentes foram isolados de compostagem por procedimentos de duas etapas. Inicialmente, as amostras tomadas em diferentes períodos e temperaturas (33 dias a 52 ºC, 60 dias a 63 ºC e mais de 365 dias a 26 ºC) foram pré-incubadas a 80 oC por 30 minutos. Posteriormente, a seleção microbiana foi conduzida por métodos baseados em cultura in vitro e choque térmico a 60 oC e 100 oC por 2h e 4h. Quarenta e um isolados foram capazes de crescer a 60 °C por 4h; vinte e sete a 100 °C por 2h e dois a 100 °C por 4h. A identificação molecular por sequenciamento parcial do gene ribossomal 16S usando primers universais revelou que trinta e cinco isolados eram de oito espécies de Bacillus, um Brevibacillus borstelensis, três Streptomyces thermogriseus e dois fungos (Thermomyces lanuginosus e T. dupontii). Os dados dos ensaios de atividade de amilase, fitase e celulase e o índice enzimático (IE) mostraram que 38 dos 41 isolados termorresistentes produziram pelo menos uma enzima. Para a atividade da amilase, o maior valor de IE foi observado em Bacillus licheniformis (isolado 21C2, IE = 4,11), seguido por Brevibacillus borstelensis (isolado 6C2, IE = 3,66), Bacillus cereus (isolado 18C2, IE = 3,52) e Bacillus paralicheniformis (isolado 20C2, IE = 3,34). Para a fitase, os maiores valores de IE foram observados para B. cereus (isolado 18C2, IE = 2,30) e B. licheniformis (isolado 3C1, IE = 2,15). ABSTRACT

RESUMEN El síndrome de shock exfoliativo(SSE) asociado a COVID- 19 es una enfermedad recientemente descrita con características superpuestas de síndrome de shock tóxico (SST) y síndrome de la piel escaldada estafilocócica (SSSS, por sus siglas en inglés). Clínicamente se presenta con fiebre, hipotensión y erupción cutánea exfoliativa.Se expone un caso en donde las lesiones cutáneas por COVID-19 precedieron a las manifestaciones respiratorias.


ABSTRACT The exfoliative shock syndrome (ESS) associated with COVID-19 is a recently described disease with overlapping features of toxic shock syndrome (TSS) and staphylococcal scalded skin syndrome(SSSS). Clinically it presents with fever, hypotension, and exfoliative skin rash. A case is presented in which the skin lesions by COVID-19 preceded the respiratory manifestations.

RESUMO A cada dia, cepas bacterianas estão tornando-se resistentes a diversos antibióticos, o que faz necessária a busca de novas substâncias eficazes para o tratamento de doenças. Desta forma, este trabalho reporta o estudo preliminar toxicológico, antibacteriano e fitoquímico do extrato etanólico das folhas de Jatropha mollissima (pinhão-bravo, Euphorbiaceae), coletada no Município de Tauá, Ceará, Nordeste Brasileiro. Inicialmente, realizou-se o teste de toxicidade do extrato contra Artemia salina. Na sequencia, foi realizado o ensaio antibacteriano contra quatro cepas bacterianas Gram-negativas (Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853, Hafnia alvei ATCC 51873, Klebsiella pneumoniae ATCC 13883) e uma cepa Gram-positiva (Enterococcus faecalis ATCC 29212). Finalmente, fez-se a análise fitoquímica preliminar do extrato ativo para detecção das principais classes de metabólitos especiais. Como resultado, o extrato etanólico das folhas de J. mollissima se mostrou tóxico para Artemia salina, pois apresentou CL50 igual a 406,02 μg/mL. Quanto à ação antibacteriana, o extrato se mostrou ativo contra a bactéria Gram-positiva Enterococcus faecalis ATCC 29212, apresentando moderada atividade antibacteriana (halo de inibição igual a 7,03 mm). Evidenciou-se no extrato bioativo a presença de cumarinas, fenóis, taninos, flavonoides (flavonóis e flavanonas), alcaloides e esteroides, ambas as classes reportadas como antimicrobianos. Portanto, esse extrato tem potencial para ser usado na produção de fármacos contra infecções causadas por bactérias Gram-positivas. No entanto, as informações direcionam estudos futuros para o isolamento e identificação dos compostos bioativos, monitorados sob a ação antibacteriana mais expressiva.


ABSTRACT Each day, bacterial strains are becoming more resistant to various antibiotics, which requires the search for new effective substances for the treatment of diseases. Thus, this study reports the toxicological, antibacterial, and phytochemical preliminary study of the ethanolic extracts of Jatropha mollissima (pinhão-bravo, Euphorbiaceae) leaves, collected in Tauá, Ceará, Northeast of Brazil. Initially, we performed the toxicity testing of the extract against Artemia salina. Then, we conducted the antibacterial assay against four Gram-negative bacterial strains (Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853, Hafnia alvei ATCC 51873, Klebsiella pneumoniae ATCC 13883), and one Gram-positive strain (Enterococcus faecalis ATCC 29212). Finally, we carried out the preliminary phytochemical analysis of the active extract to detect the main classes of special metabolites. As a result, the ethanolic extract of J. mollissima leaves was toxic to Artemia salina, because it presented LC50 equal to 406.02 µg/mL. Regarding antibacterial action, the extract was active against the Gram-positive bacteria Enterococcus faecalis ATCC 29212, with moderate antibacterial activity (inhibition zone equal to 7.03 mm). The bioactive extract had the presence of coumarins, phenols, tannins, flavonoids (flavanols and flavonones), alkaloids and steroids, both classes reported as antimicrobials. Therefore, this extract has the potential to be used in the production of drugs against infections caused by Gram-positive bacteria. However, these information require further studies for the isolation and identification of bioactive compounds, monitored under the more expressive antibacterial action.

Analisou-se a evolução anual da qualidade de leite cru refrigerado pelo levantamento e análise estatística do banco de dados das análises individuais de leite dos tanques refrigeradores, computadas mensalmente entre abril de 2002 e dezembro de 2008 em uma indústria de Minas Gerais. O volume de leite em conformidade com a IN51 para contagem bacteriana total para o ano de 2008 aumentou de 74,3% em 2002 para 85,8% em 2008. Considerando-se a IN51 para o ano de 2011, esses valores seriam de 30,5% e 46,6%, respectivamente. O volume de leite em conformidade com a IN51 para contagem de células somáticas reduziu-se de 93,5% em 2002 para 90,1% em 2008. Para o ano de 2011, esses valores seriam de 64,9 e 60,5%, respectivamente. Em relação à composição sólida do leite, o volume de leite em conformidade com a IN51 para os teores de gordura, de proteína e de extrato seco desengordurado foi de 95,2%, 98,2% e 89,6%, respectivamente. As variações sazonais influenciaram a qualidade do leite cru refrigerado de tanques de expansão. A implantação de um sistema de pagamento por qualidade do leite ao produtor foi eficaz na melhoria dos índices de qualidade da matéria-prima.


The present study aimed to analyze the annual evolution of cooled raw milk quality, through analytical analysis of the data base of individual bulk milk tanks analysis, monthly computed from April/2002 to December/2008 at a dairy industry in Minas Gerais. The mean volume of milk in compliance with IN51 for total bacterial count for year 2008 increased from 74.3% in 2002 to 85.8% in 2008. Considering IN51 for year 2011, these values would be 30.5% and 46.6%, respectively. The mean volume of milk in compliance with IN51 for somatic cells count reduced from 93.5% in 2002 to 90.1% in 2008. Considering the legal standard for the year of 2011, these values would be 64.9 and 60.5%, respectively. In relation to the solid composition of milk, the mean volume of milk in compliance with IN51 for fat, protein and solids nonfat were 95.2%, 98.2% and 89.6%, respectively. The season variations affected the quality of the bulk tank cooled raw milk from bulk tanks. The implantation of a payment system for the quality of milk to the producer revealed to be efficient in its objective to improve the quality indexes of raw milk.

ABSTRACT

Healthy term newborn feeding choice at the Maternity Ward may determine exclusive breastfeeding (EB) duration. Objective: Determine prospectively if early introduction of breast milk substitutes (BMS) or other liquids in healthy term newborns is associated to a reduction of EB incidence at 1 month of age. Methods: Concurrent cohort study of 211 healthy term neonates born in Hospital Clínico Universidad Católica during October 2007-June 2007, 108 non-supplemented and 103 supplemented with milk formula or dextrose 5 percent during the time they stayed at the Maternity Ward. Crude and adjusted risk of EB cessation between both groups was estimated at 1 month of age. Results: One month after birth, 40/100 (40.0 percent) of supplemented newborns and 26/101 (25.7 percent) non-supplemented newborns were not receiving exclusive breastfeeding (crude RR =1.55 (CI95 percent 1.03-2.34)). Adjusted by maternal characteristics (age, education, breastfeeding experience, education in breastfeeding, EB time projection and paternal support) and newborn characteristics (type of delivery and birth weight), EB cessation risk was 55 percent higher in supplemented neonates (adjusted RR = 1.55, CI95 percent 1.01-2.35). Conclusion: Early supplementation in healthy term newborns with BMS is associated to a higher risk of EB cessation at 1 month of age.


Introducción: La forma de alimentación de un recién nacido de término (RNT) durante su estadía en la Maternidad puede condicionar la duración de la lactancia materna exclusiva (LME). Objetivo: Determinar prospectivamente si la introducción precoz de SLM (sustitutos de lactancia materna) u otros líquidos a RNT sanos se asocia a una reducción de la incidencia de LME al mes de vida. Métodos: Estudio de cohorte concurrente. Se conformó una cohorte de 211 RNT sanos cuyo parto fue atendido en la Maternidad del Hospital Clínico Universidad Católica en el período octubre 2006-junio 2007, 108 no-suplementados y 103 suplementados con fórmula láctea o suero glucosado al 5 por ciento durante su estadía en la Maternidad. Se estimó el riesgo crudo de cesación de LME al mes de vida entre los grupos no-suplementado vs suplementado, y ajustado por diversas variables confundentes. Resultados: Transcurrido un mes de vida, 40/100 (40,0 por ciento) recién nacidos suplementados y 26/101 (25,7 por ciento) recién nacidos no-suplementados habían dejado de recibir LME, siendo el RR crudo = 1,55 (IC95 por ciento 1,03-2,34). Ajustado por características maternas (edad, educación, experiencia previa de lactancia, educación en lactancia, proyección LME y apoyo paterno) y del recién nacido (vía de parto y peso de nacimiento), el riesgo de cesación de LME al mes de vida fue 55 por ciento mayor en los RNT suplementados vs. no-suplementados (RR ajustado = 1,54; IC95 por ciento 1,01-2,35). Conclusión: La suplementación precoz de RNT sanos con SLM asocia a un mayor riesgo de cese de LME al mes de vida.

ABSTRACT

Dry eye is a multifactorial condition that results in a dysfunctional lacrimal functional unit. Evidence suggests that inflammation is involved in the pathogenesis of the disease. Changes in tear composition including increased cytokines, chemokines, metalloproteinases and the number of T cells in the conjunctiva are found in dry eye patients and in animal models. This inflammation is responsible in part for the irritation symptoms, ocular surface epithelial disease, and altered corneal epithelial barrier function in dry eye. There are several anti-inflammatory therapies for dry eye that target one or more of the inflammatory mediators/pathways that have been identified and are discussed in detail.


Olho seco é uma doença multifatorial que resulta em disfunção da unidade lacrimal glandular. Evidências sugerem que inflamação está involvida na patogênese da doença. Mudanças na composição das lágrimas, incluindo aumento de citocinas, quimiocinas, metaloproteinases e o número de células T na conjuntiva são encontrados em pacientes com olho seco e em modelos animais. Esta inflamação é responsável em parte pelos sintomas de irritação, doença epitelial de surperfície ocular e função epitelial de barreira alterada em olho seco. Existem várias terapias antiinflamatórias que se direcionam para um ou mais mediadores/vias que foram identificados e são discutidos em detalhe.

ABSTRACT

El acné es una patología que afecta casi al 80% de la población en algún momento de la vida. Múltiples terapéuticas locales y generales han sido utilizadas. En el presente estudio evaluamos la eficacia y seguridad del tratamiento de las lesiones activas de acné con luz pulsada como única terapéutica. Fueron tratados 91 pacientes y los resultados indican que la fototerapia es una opción terapéutica local con pocos efectos colaterales que evitaría el uso de drogas por vía general. Estos resultados fueron obtenidos en un período promedio de 4 semanas


Acne is a very common disease which affects a lot of people around the world. Multiple treatments options are available nowadays. In this study we evaluate the efficacy of IPL in active lesions of acne, as unique therapy. We treated 91 patients and results show the reduction of lesions in periods of follow-up between 4 to 6 weeks and provide a non-invasive alternative with drugs with potential secondary effects

La lipomatosis de Madelung o Lipomatosis simétrica múltiple, es una enfermedad poco frecuente, caracterizada por la presencia de lipomas no encapsulados, localizados en cuello y mitad superior de tronco. La patogenia estaría relacionada con un defecto a nivel de las catecolaminas que inducen la lipólisis. Esta patología la encontramos frecuentemente asociada al alcoholismo, hepatopatías y tumores malignos. En este artículo presentamos un caso clásico de lipomatosis de Madelung y una revisión del tema

ABSTRACT

The nutritional condition of elderly persons has close relations with their immunologic status and general health. There is no agreement regarding the recommended dietary allowances of energy and of the various nutrients inthe elderly population. Also, studies on this field were carried out in developed Countries, with possibly different cultural, social, and even climatic conditions if compared with Brazil. This paper revises some aspects of the energy and protein metabolism in elderly persons and highlights the necessity for new research lines about the nutritional status and the adequacy of the caloric and protein intake of our aged population in order to enable the development of rational nutritional interventions


El estado nutricional de lo adultos mayores influye en su sistema inmunológico y la salud. No existe un consenso acerca de las necesidades calóricas y de nutrientes para esta población. La mayor parte de los estudios de estas variables fueron realizados en países desarrollados, con condiciones culturales, sociales y hasta climáticas, bastante diferentes del Brasil. Se revisaron algunos aspectos del metabolismo enegético y proteico de los adultos mayores, señalando los estudios necesários para uma evaluación del estado nutricional, que permitan el ajuste correcto de estos parámetros en la dieta de los adultos mayores, elementos indispensables para la planificación de intervenciones nutricionales racionales


O estado nutricional dos idosos é relacionado com o seu estado imunológico e saúde geral. Não existe consenso sobre as necessidades calóricas e dos diversos nutrientes na população idosa. Também, a maioria dos estudos acerca destas variáveis foram realizados em países desenvolvidos, com cultura, condições sociais e mesmo climáticas diferentes daquelas do Brasil. ABSTRACT

1. Two types of nylon-6 supports (small cylinders and a sponge-like derivative) were prepared for immobilizing enzymes. Nylon-6 beads were solubilized by immersion in 80 formic acid and then reprecipitated using two different types of non-solvent solutions (distilled water or a 1:1 acetone:water solution) giving rise to a sponge-like derivative and to a colloidal suspension, respectively. The latter was molded into a thin thread which was cut into small cylinders. 2. Trypsin (EC 3.4.21.4) was covalently bound to glutaraldehyde-activated nylon-6 cylinders as well as to the sponge-like derivative. The maximum (100) apparent initial enzymatic activity was found for the trypsin bound to small cylinders, while the initial activity of trypsin bound to the sponge-like material was 61 in comparison with that of trypsin-small cylinders, under the same conditions of enzyme immobilization reaction (1 g of nylon support and 5 ml of 1.3 mg/ml trypsin in 0.1 M sodium phosphate buffer, pH 8.5, at 10 degrees C for 18 h) and of enzymatic reaction (1 g of trypsin-nylon in a batch reactor, 2 ml of 0.7 w/v azocasein solution in 50 mM borate buffer, pH 8.5, at 37 degrees C, with shaking, for 1 h). However, the decrease of activity after enzyme immobilization was more conspicuous for the trypsin-small cylinders than for the trypsin-sponge. The former retained approximately 25 of its initial activity, while the latter retained approximately 67 of its initial activity, after seven cycles of utilization for 1 h, pH 8.5, at 37 degrees C and 8 days of storage, pH 8.5, at 4 degrees C in the presence of azocasein. 3. Scanning electron microscopy was performed to visualize the surface of the support after each step of the immobilization process. The electron micrographs show that the two types of nylon supports had a rough surface, which became rougher and full of craters after treatment with 5 N HCl. On the other hand, the partially hydrolyzed nylon surface acquired the appearance of Swiss cheese after treatment with 2.5 glutaraldehyde. After reaction with the enzyme molecules the surface became rougher again.

ABSTRACT

1. Trehalase was partially purified from Escherichia coli and characterized. The Km for trehalose was 0.78 mM, the pH optimum 5.5 and the temperature optimum 30 degrees C. 2. Trehalase represented approximately 50 per cent of the total protein released by osmotic shock. The preparation was free of nonspecific carbohydrate hydrolases, which act on sucrose, galactose and maltose, permitting trehalose determination in biological samples, such as insect hemolymph and free cell extracts among others. 3. The enzyme was stable in 50 mM maleate buffer, pH 6.2, at -8 degrees C for at least 6 months and could be used to determine trehalose in the range of 6 to 30 nmol. 4. Immobilization of the enzyme was achieved by covalent linkage to spherisorb-5NH2 (spherical silica gel). Retention of total catalytic activity averaged 32 per cent . 5. The reactor, stored for one month at -5 degrees C, retained 98 per cent of its initial immobilized activity. 6. This immobilized form of the enzyme could be used routinely for specific determinations of trehalose
